What is RPPA data?

The reverse phase protein array (RPPA) data platform provides expression data for a prespecified set of proteins, across a set of tissue or cell line samples. RPPA data can be combined with those of other molecular profiling platforms, in order to obtain a more complete molecular picture of the cell.

What is RPPA used for?

RPPA represents an antibody‐based functional proteomic analysis for both tumor tissue and cultured cells. RPPA characterizes the basal protein expression and modification levels, growth factor‐ or ligand‐induced effects, and time‐resolved responses appropriate for systems biology analysis.

What is proteomic testing?

Proteomic testing (VeriStrat®) is proposed as a blood-based predictive and prognostic test for individuals with non-small-cell lung cancer (NSCLC) who test negative for EGFR mutations (EGFR wild-type) or whose EGFR mutation status is unknown and for individuals who have not responded to first line therapies.

How many reads are needed?

Most experiments require 5–200 million reads per sample, depending on organism complexity and size, along with project aims. Gene expression profiling experiments that are looking for a quick snapshot of highly expressed genes may only need 5–25 million reads per sample.

Why do you need methanol in transfer buffer?

The presence of methanol in the transfer buffer serves two main purposes: It promotes dissociation of SDS from the protein and dramatically improves adsorption of proteins onto membranes in the presence of SDS, although these effects may vary with proteins.

Do Western blots use antibodies?

Antibodies are used to detect target proteins on the western blot (immunoblot). The antibodies are conjugated with fluorescent or radioactive labels or enzymes that give a subsequent reaction with an applied reagent, leading to a coloring or emission of light, enabling detection.

How is RPPA used in the treatment of cancer?

Studying large-scale epigenetic changes such as protein phosphorylation or cleavage will greatly aid in understanding the causes and determining effective treatment of cancers and other complex diseases. RPPA represents an antibody‐based functional proteomic analysis for both tumor tissue and cultured cells.

How is reverse phase protein array ( RPPA ) used?

Reverse-phase protein array (RPPA) is a high-throughput antibody-based targeted proteomics platform that can quantify hundreds of proteins in thousands of samples derived from tissue or cell lysates, serum, plasma, or other body fluids. Protein samples are robotically arrayed as microspots on nitroc …

Why is MD Anderson’s RPPA core so important?

MD Anderson RPPA Core is a valuable resource to the scientific community due to its unique and continuous validation and quality control of all aspects of its RPPA Pipeline, including the below: Serial dilutions of each sample to capture the linear antibody/antigen reaction for accurate data analysis.
